Madekwe is the daughter of an ethnically diverse family in South London in England. Madekwe is English as well as her father was Nigerian. She went to The Brit School when she was 14. She graduated with a BA in acting at The Royal Academy of Dramatic Art. Madekwe had her official US debut on the drama series The Beautiful Life, which just aired two episodes on The CW before being cancelled. Ashley Davenport, a character in ABC's Revenge drama, was chosen for her as Ashley Davenport. Madekwe was a regular on the drama series of Revenge on ABC for two seasons. The show was announced in May 2013 that she would leave the series after making an appearance in the first season of the third. Between 2014 and 2016, she starred in Salem on the CW as Tituba. Madekwe wed her actor boyfriend Iddo goldberg on June 2012. They've collaborated in the show Secret Diary of a Call Girl and he also had a role alongside her in the television series Salem.
